Complement C1q A Chain (C1qa) is a subunit of C1q. C1q is composed of 6 polypeptide chains each of C1qa, C1qb, and C1qc. C1q is a component of the classical complement pathway, and thereby plays an important role in the immune response. Each C1q subunit contains an N-terminal collagen-like region and a C-terminal C1q globular domain. The presence of receptors for C1q on effector cells modulates its activity, which may be antibody-dependent or independent. Macrophages are the primary source of C1q, while anti-inflammatory drugs as well as cytokines differentially regulate expression of the mRNA as well as the protein. This antibody recognizes a protein of ~35 kDa, identified as CD74. It is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. This MAb recognizes a protein of 52 kDa, which is identified as TNFR1 (TNFR-associated factor 1). CD30-positive lymphoproliferations of the skin comprise 30% of all primary cutaneous T-cell lymphomas (CTCLs). Besides borderline cases this group includes lymphomatoid papulosis (LyP) and primary cutaneous anaplastic large T-cell lymphoma (cALCL). Although the two entities overlap clinically, histopathologically, immunopathologically and genetically, they differ considerably in their prognosis. In particular, common feature of both cases is histologically the presence of atypical lymphoid CD30-positive T blasts and genetically a clonal T-cell-receptor rearrangement. However, both cases differ considerably in their clinical course: Lesions of LyP regress spontaneously, whereas those of cALCL persist and may progress and spread. This MAb recognizes a 21 kDa protein, identified as the p21WAF1 tumor suppressor protein. This MAb is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk's and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53. Its expression is induced by the wild type, but not mutant, p53 suppressor protein. Normal cells generally display a rather intense nuclear p21 expression. Gastrin, is a hormone that normally formed by mucosal cells in the gastric antrum and by the D cells of the pancreatic islets. Its primary function is to stimulate secretion of HCl by the gastric mucosa. HCl, in turn, inhibits gastrin formation. It also responsible for stimulating smooth muscle contraction and increasing blood circulation and water secretion in the stomach and intestine. Gastrin is regulated by epidermal growth factor in both mice and humans. Gastrin is excreted in excess by pancreatic tumors in the Zollinger-Ellison syndrome. Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. The finding that mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC) has resulted in considerable interest in the understanding of the mechanism of DNA mismatch repair. Initially, inherited mutations in the MSH2 and MLH1 homologs of the bacterial DNA mismatch repair genes mutS and mutL were demonstrated at high frequency in HNPCC and were shown to be associated with microsatellite instability. Band 3, also designated AE1, is an erythrocyte membrane glycoprotein that contributes to cell stuctural integrity and mediates exchange of chloride and bicarbonate across the phospholipid bilayer. The diverse functions of the approximately 900 amino acid protein are mediated by two distinct domains. The amino-terminal domain, also known as cdb3 for cytoplasmic domain of erthrocyte membrane band 3, acts as an attachment site for the erythrocyte skeleton by binding ankyrin. The carboxy-terminal, membrane-associated domain carries out exchange transport of anions. Degradation of band 3 can generate an aging antigen known as senescent cell antigen, or SCA, which is expressed on old cells and marks them for removal by the immune system. This monoclonal antibody is specific to estrogen receptor alpha (ER alpha) and shows minimal cross-reaction with other members of the family. ER is an important regulator of growth and differentiation in the mammary gland.
